Abstract

Objective: To evaluate the levels of IFN-γ and IFN-γR1 in trophoblastic tissue of women with Toxoplasma gondii.

Methods: This study included 126 women with spontaneous abortion. Serum was obtained for performance of ELISA test for detection of anti-Toxoplasma IgM. Paraffin embedded blocks of trophoblastic tissue obtained from each patient were prepared and investigated for presence of T. gondii antigen, IFN-γ and IFN-γR1 using immunohistochemical analysis.

Results: The frequency of the T. gondii infection was high among women with abortion, 23 of 120 women (19.17%) have IgM Abs against T. gondii. The majority of patients within the positive and negative groups were found to have no previous abortions, while patients with previous abortions constituted a less percent. Among the induced abortion group, it was found that all six cases had no previous abortions. There was a highly significant difference in mean percent of IFN-γ and IFN-γR1 when compared between positive and negative groups, while there was no such significant difference regarding mean percent of IFN-γR1/ IFN-γ ratio between positive and negative groups. There was a highly significant difference regarding mean percent of IFN-γ and IFN-γR1 and a significant difference regarding mean percent of IFN-γR1/ IFN-γ ratio between positive and induced abortion groups. Similarly, a significant difference was found when mean percent of IFN-γ, IFN-γR1 and IFN-γR1/ IFN-γ ratio were compared between negative and induced abortion groups. Ratio of IFN-γ R1 to IFN-γ was 1.2:1 in positive group and 1.28:1 in negative group, while in induced abortion group it was 2:1.

Keywords: Toxoplasma gondii, Interferon-γ, interferon–γ receptor, spontaneous abortion.
